Normothermic machine perfusion (NMP) is nowadays frequently utilized in liver transplantation. Despite commonly accepted viability assessment criteria, such as perfusate lactate and perfusate pH, there is a lack of predictive organ evaluation strategies to ensure graft viability. Hyperspectral imaging (HSI)-as an optical imaging modality increasingly applied in the biomedical field-might provide additional useful data regarding allograft viability and performance of liver grafts during NMP. Methods: Twenty-five deceased donor liver allografts were included in the study. During NMP, graft viability was assessed conventionally and by means of HSI. Images of liver parenchyma were acquired at 1, 2, and 4 h of NMP, and subsequently analyzed using a specialized HSI acquisition software to compute oxygen saturation, tissue hemoglobin index, near-infrared perfusion index, and tissue water index. To analyze the association between HSI parameters and perfusate lactate as well as perfusate pH, we performed simple linear regression analysis. Results: Perfusate lactate at 1, 2, and 4 h NMP was 1.5 [0.3-8.1], 0.9 [0.3-2.8], and 0.9 [0.1-2.2] mmol/L. Perfusate pH at 1, 2, and 4 h NMP was 7.329 [7.013-7.510], 7.318 [7.081-7.472], and 7.265 [6.967-7.462], respectively. Oxygen saturation predicted perfusate lactate at 1 and 2 h NMP (R2 = 0.1577, P = 0.0493; R2 = 0.1831, P = 0.0329; respectively). Tissue hemoglobin index predicted perfusate lactate at 1, 2, and 4 h NMP (R2 = 0.1916, P = 0.0286; R2 = 0.2900, P = 0.0055; R2 = 0.2453, P = 0.0139; respectively). Conclusions: HSI may serve as a noninvasive tool for viability assessment during NMP. Further evaluation and validation of HSI parameters are warranted in larger sample sizes.

Good adaptive support by educational professionals is crucial for pre-school children's mathematical learning. Thus there is a need for appropriate instruments to evaluate the quality of the planning, implementation, and reflection process for mathematical learning opportunities in kindergarten. Existing instruments tend to focus on micro-adaptive learning support (MI-ALS), which considers the interaction between the professionals and the children. The quality of macro-adaptive learning support (MA-ALS), the teachers' planning of and reflection on learning opportunities, has yet to be comprehensively investigated. MA-ALS is important for the acquisition of subject-related (mathematical) competence, an aspect of kindergarten education that is gaining in importance in the pre-school curricula of many countries.The quality of mathematical learning support in kindergarten is conceptualized by differentiating between the quality of MA-ALS and MI-ALS. MI-ALS is also divided into general support (group management, emotional warmth) and subject-related support (learning stimulation, subject-specific language). A rating tool for analyzing the quality of mathematical learning support is presented and its psychometric quality is assessed. The tool is then used to analyze the teaching quality in two guided play situations (recorded) and two interviews each with 145 education professionals in Germany and Switzerland. A confirmatory factor analysis (CFA) confirms the validity of the distinction between group management, emotional warmth and subject-related support. The relationship between the level of MI-ALS and MA-ALS, the training of the teachers (academic vs non-academic), and the educational context (Germany vs Switzerland) is examined. The results corroborate the importance of including planning and reflection as a dimension of learning support quality.

Mannosylerythritol lipids are glycolipid biosurfactants with many interesting properties. Despite the general interest in those molecules and the need for a robust process, studies on their production in bioreactors are still scarce. In the current study, the fermentative production of MEL in a bioreactor with Moesziomyces aphidis was performed using a defined mineral salt medium. Several kinetic process parameters like substrate consumption rates and product formation rates were evaluated and subsequently enhanced by increasing the biomass concentration through an exponential fed-batch strategy. The fed-batch approaches resulted in two to three fold increased dry biomass concentrations of 10.9-15.5 g/L at the end of the growth phase, compared with 4.2 g/L in the batch process. Consequently, MEL formation rates were increased from 0.1 g/Lh up to around 0.4 g/Lh during the MEL production phase. Thus, a maximum concentration of up to 50.5 g/L MEL was obtained when oil was added in excess, but high concentrations of residual fatty acids were also present in the broth. By adjusting the oil feeding to biomass-specific hydrolysis and MEL production rates, a slightly lower MEL concentration of 34.3 g/L was obtained after 170 h, but at the same time a very pure crude lipid extract with more than 90% MEL and a much lower concentration of remaining fatty acids. With rapeseed oil as substrate, the ideal oil-to-biomass ratio for full substrate conversion was found to be around 10 goil/gbiomass. In addition, off-gas analysis and pH trends could be used to assess biomass growth and MEL production. Finally, kinetic models were developed and compared to the experimental data, allowing for a detailed prediction of the process behavior in future experiments.

INTRODUCTION: In liver transplantation (LT), steatosis is commonly judged to be a risk factor for graft dysfunction, and quantitative assessment of hepatic steatosis remains crucial. Liver biopsy as the gold standard for evaluation of hepatic steatosis has certain drawbacks, that is, invasiveness, and intra- and inter-observer variability. A non-invasive, quantitative modality could replace liver biopsy and eliminate these disadvantages, but has not yet been evaluated in human LT. METHODS: We performed a pilot study to evaluate the feasibility and accuracy of hyperspectral imaging (HSI) in the assessment of hepatic steatosis of human liver allografts for transplantation. Thirteen deceased donor liver allografts were included in the study. The degree of steatosis was assessed by means of conventional liver biopsy as well as HSI, performed at the end of back-table preparation, during normothermic machine perfusion (NMP), and after reperfusion in the recipient. RESULTS: Organ donors were 51 [30-83] years old, and 61.5% were male. Donor body mass index was 24.2 [16.5-38.0] kg/m2 . The tissue lipid index (TLI) generated by HSI at the end of back-table preparation correlated significantly with the histopathologically assessed degree of overall hepatic steatosis (R2 = .9085, P < .0001); this was based on a correlation of TLI and microvesicular steatosis (R2 = .8120; P < .0001). There is also a linear relationship between the histopathologically assessed degree of overall steatosis and TLI during NMP (R2 = .5646; P = .0031) as well as TLI after reperfusion (R2 = .6562; P = .0008). CONCLUSION: HSI may safely be applied for accurate assessment of hepatic steatosis in human liver grafts. Certainly, TLI needs further assessment and validation in larger sample sizes.

This study demonstrates the application and combination of multiple imaging techniques [light microscopy, micro-X-ray computer tomography (µ-CT), scanning electron microscopy (SEM) and focussed ion beam - nano-tomography (FIB-nT)] to the analysis of the microstructure of hydrated alite across multiple scales. However, by comparing findings with mercury intrusion porosimetry (MIP), it becomes obvious that the imaged 3D volumes and 2D images do not sufficiently overlap at certain scales to allow a continuous quantification of the pore size distribution (PSD). This can be overcome by improving the resolution and increasing the measured volume. Furthermore, results show that the fibrous morphology of calcium-silicate-hydrates (C-S-H) phases is preserved during FIB-nT. This is a requirement for characterisation of nano-scale porosity. Finally, it was proven that the combination of FIB-nT with energy-dispersive X-ray spectroscopy (EDX) data facilitates the phase segmentation of a 11 × 11 × 7.7 µm3 volume of hydrated alite.

Here, we report the complete genome sequence of Bacillus velezensis strain S4, which was isolated from biochar-amended agricultural soil collected in Smyrna, Delaware. The genome is 4.07 Mbp, encodes 3,918 predicted proteins, and has a GC content of 46.4%.
